For instance, if I have a class like:

.class1 {
   font-family: Arial, Serif;
}

...and I actually have Arial on my computer, but I wish to test out the site using Serif (without changing the code for the class), the plugin would make the browser pretend that the font-family Arial doesn't exist, and display Serif instead.

No plugins that I know of, but you could always do this dynamically in Firebug when you hit the page. You can just go to that style, and set it to be serif instead of Arial, serif.
